  Nefiracetam exerts a number of different effects onseveral neurotransmitter systems in the brain. Like other racetamcompounds, it has been shown to modulate acetylcholineneurotransmission by accelerating turnover and release. Unlikeother racetam compounds, it can bind to GABA receptors. Thisinteraction between nefiracetam and GABA receptors is believed tohave favorable effects on memory consolidation. Nefiracetam mayalso augment ion flow into neurons through NMDA receptors that arenear cholinergic (acetylcholine) receptors. NMDA receptors aresubtype of ion channel that are essential for learning and memory.It also increases levels of enzymes needed to synthesizenoradrenaline, dopamine, and serotonin in the hippocampus andfrontal cortex.Strong anti-epileptic effects have also beenobserved in mice and rats, but the mechanism behind this remainsunknown. Nefiracetam has also been used to increase blood flowto the brain of rats with occluded carotidarteries. Nefiracetam BenefitsStudies in mice havedemonstrated that nefiracetam significantly improves spatialmemory; mice given the drug do better in mazes and on recognitiontasks. Damage from insults such as ischemia (restricted bloodsupply/low oxygen) or anoxia (no oxygen) is reduced in animals thathave been treated with nefiracetam. It has also been shown toimprove learning in a rat model of Alzheimer’s disease, andclinical studies of nefiracetam for Alzheimer’s patients areongoing. One study showed that nefiracetam prevented amnesiainduced by propofol (the anesthetic of Michael Jackson fame).Interestingly, the compound may also be useful to treat chronicpain; it reduces thermal hyperalgesia in mice.The effects in humansare still being investigated, but the clinical trials that havetaken place suggest that nefiracetam may improve cognitive functionand frontal lobe activity. It also has antidepressive effects;patients who had suffered strokes and had severe post-strokedepression had improved outlooks, decreased apathy, and higherenergy after taking nefiracetam for at least 4 weeks (the studylasted 12 weeks).Anecdotal benefits of nefiracetam in otherwisehealthy people include improved recall, anxiolytic effects, bettercoordination, and improved attention/focus.
