Direct random sequencing of clones from standard cDNA libraries is inefficient for discovering rare transcripts, because cDNA of medium and high abundance are sequenced repeatedly instead. Normalization performed before cDNA library sequencing decreases the prevalence of clones representing abundant transcripts.
